That depends very much are where you're flying to and from. You usually want to look for the most direct flights, with the fewest stops. Otherwise, they're pretty much all the same.

How would you define 'best'? I prefer Southwest, if they fly where I want to go, because I like to pay as little as possible.

"Best" is subjective. Do you want best price, best flight map for you, best rewards program, best bells & whistles while in flight...
And, no, Southwest is far from the best. They are good low end airline, but they are hardly the best. Any airline which has a cattle call to board and hope you get a decent seat can never be the best.
